Content of the Module:

This course presents introductory biochemistry relevant to human health and disease that forms part of the foundation of modern medical practice. The molecular correlation with disease is emphasized throughout the course.

The topics included are:

- Enzymes: introduction, nomenclature, classification, mechanism of action, inhibition and factors affecting activity.

- Vitamins, introduction, nomenclature, classification, mechanism of action, diseases states associated with defeciency.

- Carbohydrates metabolism and disease states associated with disorders of metabolism.

- Lipid metabolis and disease states associated with disorders of metabolism.

- Protein and amino acid metabolism and disease states associated with disorders of metabolism.

- Nucleic acid metabolism and disease states associated with disorders of metabolism.

• Recognize the Theoretical knowledge about microscopy (light microscopy [LM], as well as TEM and SEM).

• Describe the applications of LM, TEM, and SEM, Illumination sources (photons vs. accelerated electrons), Lens systems (glass vs. electro-magnetic), Magnification vs. resolution, Specimen-electron beam interactions.

• Compare between Electron microscope (EM) and Light microscope (LM).

• Explain the Principles and construction of TEM & SEM.

• Summarize and differentiate between specimen preparations for TEM & SEM.

• Work effectively in team.

• Perform the steps required for specimen preparation for transmission electron microscopy (TEM): Fixation and dehydration, Embedding, Ultrathin sectioning (ultramicrotomy), Staining grids with heavy metal stains.

• Introduction to electron microscopy : fundamental concepts in electron microscopy; history and pioneers in electron microscopes, the main two types of electron microscopes.

• Light microscopy vs electron microscopy.

• Comparison between Electron microscope (EM) and Light microscope (LM) :Similarities (Arrangement and function of component are similar), Differences between light and electron microscopes.

• The Scale of things and resolving power line.

• Advantages and disadvantages of EM.

• Electrons/wave length/ resolution.

• Relationship between wavelength and resolution: Relationship between wavelength of electron and resolution ,Relationship between electron wavelength and accelerating voltage(de Broglie equation) and Intermediate and high voltage EM (ADVANTEGES AND EXAMPLES).

• Magnification vs. resolution.

• Optics vs electromagnetic lenses, and defect of lenses(lens aberrations).

• Interactions between specimen and electron beam.

• Principles and construction of transmission electron microscope (TEM).

• Specimen preparation for transmission electron microscopy (TEM): Fixation and dehydration, Embedding, Ultramicrotomy (sectioning by ultra-microtome).

• Staining methods : staining of ultra-thin sections by heavy metals stains and contrast and image formation for TEM, Negative staining.
